DJ Hero è il primo titolo della collana di videogiochi musicali della Activision dedicato al mondo dei DJ. Sviluppato da FreeStyleGames e pubblicato perlappunto da Activision - come tutti i titoli del "fratellone rock" Guitar Hero - è stato pubblicato il 27 ottobre 2009 in Nord America e il 29 ottobre 2009 in Europa. Il gioco è basato sul mix di brani musicali con stile e caratteristiche che si avvicinano ai ritmi della Disco anni 70/80 e dell'Hip-hop/RNB. Per giocare l'utente deve premere i 3 pulsanti colorati (verde,rosso e blu) ed adeguare la loro dissolvenza "Scratchando" il giradischi sul controller del gioco per guadagnare punti. Il gioco è dotato di modalità single player, Carriera, cooperativo e multiplayer.
In alcuni brani il gioco offre anche la possibilità di connettere il proprio controller chitarra o il microfono per formare una band con i propri amici.
2 Many DJs e tanti artisti mix hanno contribuito al gioco in fase di sviluppo del gioco, nella creazione dei mix e per gli avatar giocabili.
Il controller è costituito da un ponte wireless costituito, come accennato poc'anzi, da un giradischi che supporta 3 "stream" pulsanti con linea effetti, un crossfader ed un "Euphoria" button. In aggiunta al mix vi è un pannello nascosto con dei pulsanti di controllo ulteriori che serviranno per interagire con la console di gioco.
Come per le versioni con chitarra, anche Dj Hero può essere configurato per giocatori mancini.
Il gioco si presenta così:

il giocatore deve tenere premuto due pulsanti che riflettono i due brani utilizzati in quella particolare miscela, mentre il terzo rappresenta i campionamenti da aggiungere che possono essere regolati con la manopola effetti.
Il giocatore deve anche costantemente regolare il crossfader per migliorare il suono della canzone, ottendo così una maggiore quantità di punti.
Alcuni brani vengono visualizzati sullo schermo come una serie di frecce indicanti su o giù rappresentando le sezioni "graffi", vale a dire la direzione verso la quale il giocatore deve fa "girare" il disco proprio come un vero disc-jokey. L'"Euphoria" button è l'equivalente dello Star Power di Guitar Hero e può essere rilasciato premendo il pulsante Euphoria, il tutto per raddoppiare i punti ottenuti.
Dieci canzoni sono state appositamente create per supportare il gioco con le chitarre controller compatibili per vivere l'esperienza di un "DJ vs Guitar" mode.
Una modalità Jukebox permette al gioco di riprodurre automaticamente le canzoni con la possibilità di saltare e giocare con le proprie canzoni preferite in qualsiasi momento. 
La Casa distributrice del gioco ha già fatto sapere che a breve sarà possibile scaricare dei brani aggiuntivi per tutte le piattaforme di gioco.
I brani presenti nella track-list accontenteranno non solo gli appassionati di Hip-pop, ma anche cultori di altri generi musicali come grunge, soul, R & B, techno e house.
